Is patreon replying to email support?

I sent an email requesting support 2 days ago (as creator) and haven’t heard anything. I also tried on twitter.

I really need support with one of my subscribers, guys in your experience which channel is more quick for getting support as a creator?

I don’t think you’ll find any Patreon staff here. It’s mostly peer-to-peer support.

That said, you sent a support request outside of business hours, so you basically contacted them 1 day ago. Start worrying if they won’t reply by Friday.
